Fig. 3. Involvement of intracellular Ca2+ release in the HU induced stimulation of melanin synthesis in B16 melanoma cells. The data (A) represent intracellular Ca2+ changes with time. The arrow shows the time point for addition of HU (20 μM). Dant (50 μM) and 2-APB (100 μM), inhibitors of ryanodine and IP3 receptors, respectively, were added 10 min before treatment with HU. In the data (B) and (C), quantitative changes in intracellular Ca2+ concentrations ([Ca2+]i) and melanin contents, respectively, are expressed as a fold increase compared to negative control. BAPTA/AM (1 μM), an intracellular Ca2+ chelator, was used in the experiments. In these data, each column represents the mean value of four replications with bars indicating SEM. *P<0.05 compared to negative control. #P<0.05 compared to HU alone.
